Variability in Gram staining is seen in Gram positive organisms. This is commonly seen in older cells, especially in Bacillus species. Gram non-reactive organsims are likely those that have higher lipid concentrations in thier cells walls. The counterstain used in the Gram stain procedure can effect the ability to visualize some Gram negative organism such as Legionella and some anaerobes. Use of basic fuchsin as a counterstain allows for the visualization of some of these organsims which do not stain well with safranin counterstaining.
